This combination therapy consists of atorvastatin, a small molecule HMG-CoA reductase inhibitor, and celecoxib, a selective cyclooxygenase-2 (COX-2) inhibitor. Developed by Rutgers, The State University of New Jersey, the regimen is being investigated for the treatment of prostate cancer, specifically in patients with biochemical recurrence (rising PSA levels) following local therapy such as radical prostatectomy or radiation. Preclinical research from Rutgers demonstrated that this low-dose combination exerts synergistic effects in inhibiting the progression of androgen-dependent prostate tumors to androgen independence. The Phase II clinical trial (NCT01220973) evaluated the efficacy of 20 mg atorvastatin daily combined with 200 mg celecoxib twice daily, with the primary goal of increasing PSA doubling time and delaying disease progression.
