Overview

A fixed multi-drug regimen of four repurposed small-molecule pharmaceuticals—atorvastatin, metformin, doxycycline, and mebendazole—used as an adjunctive, metabolically targeted combination therapy. The concept, often referred to as the Care Oncology Protocol, aims to inhibit cancer growth and survival by simultaneously targeting dysregulated tumor metabolism, mitochondrial function, lipid/cholesterol pathways, protein synthesis, and cytoskeletal dynamics. Mechanisms include AMPK activation and mitochondrial complex I inhibition (metformin) leading to inhibition of mTOR signaling and reduced anabolic metabolism; HMG-CoA reductase inhibition (atorvastatin) decreasing mevalonate pathway flux, prenylation, and cholesterol availability critical for oncogenic signaling; 30S ribosomal subunit binding (doxycycline) inhibiting mitochondrial protein translation and biogenesis in cancer cells and cancer stem cells; and tubulin polymerization inhibition plus glycolytic interference (mebendazole) inducing mitotic arrest and apoptosis. It is positioned as adjunctive to standard-of-care oncology treatments and is not a standalone approved cancer therapy.
