Atu027 + gemcitabine is a combination therapy investigated for advanced pancreatic carcinoma and other solid tumors. Atu027 is a liposomally formulated small interfering RNA (siRNA) therapeutic that targets and silences protein kinase N3 (PKN3) expression in vascular endothelium, thereby inhibiting tumor metastasis. Gemcitabine is a nucleoside analog used as standard chemotherapy for various cancers, including pancreatic cancer. The combination aims to leverage the anti-metastatic action of Atu027 with the cytotoxic effects of gemcitabine to improve disease control in patients with advanced or metastatic cancer[1][2][5]. Clinical trials have shown that this combination is generally well tolerated, with some improvement in progression-free survival observed when using twice-weekly dosing of Atu027 alongside standard gemcitabine regimens[1][5]. Silence Therapeutics developed Atu027; it was previously under investigation for several solid tumors but development has been discontinued[8].
