ATX-898 + fulvestrant + ribociclib is an investigational triplet combination therapy being evaluated for the treatment of advanced solid tumors, including ovarian, gynecologic, and HR+/HER2- breast cancers. The regimen combines **ATX-898**, a novel small molecule precision oncology agent developed by Antares Therapeutics (a spin-out of Scorpion Therapeutics), with two established anti-neoplastic agents: **fulvestrant** and **ribociclib**. Fulvestrant is a selective estrogen receptor degrader (SERD) that binds to and destabilizes the estrogen receptor, while ribociclib is a selective inhibitor of cyclin-dependent kinases 4 and 6 (CDK4/6), which are critical regulators of the cell cycle. By simultaneously targeting the estrogen signaling pathway and the cell cycle machinery, this combination aims to enhance therapeutic efficacy and overcome resistance mechanisms in hormone-driven malignancies. The combination is currently being studied in a Phase 1/2 open-label clinical trial (NCT06465290).
