Combination immunotherapy comprising AU-007 (an interleukin-2-binding monoclonal antibody), aldesleukin (recombinant human interleukin-2), and nivolumab (a PD-1 monoclonal antibody). AU-007 redirects endogenous IL-2 toward effector T cells and NK cells, away from T regulatory cells, by inhibiting binding to CD25 and promoting CD122/CD123 binding, thereby enhancing anti-tumor immune responses. Aldesleukin exogenously increases IL-2 levels, further boosting T cell activity. Nivolumab blocks the PD-1 receptor on T cells, reversing tumor-induced immune suppression. This triplet is under investigation for advanced solid tumors including melanoma, with early-phase studies showing favorable safety and signs of durable objective responses[1][3][9]. The regimen is developed and studied by Aulos Bioscience.
