Aumolertinib + carboplatin + pemetrexed is a combination therapy used for the treatment of advanced non-small cell lung cancer (NSCLC) with epidermal growth factor receptor (EGFR) mutations. This regimen combines a third-generation EGFR tyrosine kinase inhibitor (aumolertinib) with platinum-based chemotherapy (carboplatin) and an antifolate agent (pemetrexed). In a phase II clinical trial (NCT04646824), patients received aumolertinib 110 mg once daily plus pemetrexed (500 mg/m²) and carboplatin (area under curve = 5) once every 3 weeks for 4 cycles, followed by maintenance therapy with aumolertinib (110 mg once daily) and pemetrexed (500 mg/m² once every 4 weeks). This combination showed promising efficacy with a median progression-free survival of 28.0 months, an objective response rate of 91.2%, and a disease control rate of 100%. Aumolertinib selectively inhibits EGFR sensitizing mutations and EGFR T790M resistance mutations, while the addition of chemotherapy (carboplatin + pemetrexed) may enhance antitumor effects through complementary mechanisms.
