Aumolertinib + midazolam refers to the co-administration of two drugs: aumolertinib, a third-generation epidermal growth factor receptor (EGFR) tyrosine kinase inhibitor (TKI), and midazolam, a short-acting benzodiazepine commonly used as a probe substrate for CYP3A4 activity. This combination is being studied to evaluate pharmacokinetic interactions, specifically the effect of aumolertinib on the metabolism of midazolam, in patients with EGFR mutation-positive non-small cell lung cancer (NSCLC). Aumolertinib inhibits mutant EGFR, thereby blocking downstream signaling involved in tumor growth and survival. Midazolam acts as a gamma-aminobutyric acid (GABA) A receptor modulator, inducing sedation, anxiolysis, and amnesia. The interaction study helps determine if aumolertinib acts as an inducer or inhibitor of CYP3A4, impacting the clearance of drugs metabolized by this enzyme[1][3][5][6].
