Aura Biosciences is developing a Virus-like Drug Conjugate (VDC) for the treatment of various solid tumors that express modified heparan sulfate proteoglycans (mHSPG). The VDC platform consists of a human papillomavirus (HPV)-derived capsid, which acts as a virus-like particle (VLP) delivery vehicle, conjugated to a cytotoxic payload. These VLPs naturally and selectively bind to tumor-associated glycosaminoglycans (GAGs), specifically mHSPGs, which are uniquely upregulated on the surface of many cancer cells compared to healthy tissue. Upon binding, the VDC is designed to deliver a cytotoxic effect that may also induce pro-immunogenic cell death, potentially stimulating a systemic anti-tumor immune response. This program represents an expansion of Aura's technology beyond its lead indications in ocular oncology and bladder cancer, aiming to leverage the high specificity of the VDC platform for the systemic treatment of mHSPG-positive solid tumors.
