Overview

Autologous AML cell vaccine (CD80) is an investigational, patient-specific cancer immunotherapy designed for the treatment of acute myeloid leukemia (AML). The vaccine is produced by genetically engineering a patient's own AML blast cells, typically using lentiviral vectors, to express the co-stimulatory molecule CD80 (B7-1). AML cells often lack or downregulate CD80, allowing them to evade the immune system; by restoring its expression, the vaccine provides the necessary 'second signal' (via CD28) to activate T cells against a broad range of leukemia-associated antigens. Some versions, such as TriLeukeVax (TLV), also co-express IL-15 and IL-15Rα to further enhance the survival and cytolytic activity of anti-leukemic T cells and natural killer (NK) cells. The engineered cells are irradiated prior to administration to ensure safety while maintaining immunogenicity. It is primarily investigated for patients in complete remission who have persistent minimal residual disease (MRD).
