Autologous anti-CD19 CAR-T cells are a form of cell therapy where a patient’s own (autologous) T cells are collected, genetically modified to express a chimeric antigen receptor (CAR) that specifically recognizes the CD19 antigen (a protein expressed on B cells), and then re-infused into the patient to target and eradicate malignant B cells. The CAR construct typically contains an antibody-derived antigen-binding domain linked to intracellular T-cell activation and costimulatory domains, enabling targeted recognition and destruction of B-cell malignancies in an HLA-independent manner. This therapy is primarily used in relapsed or refractory B-cell hematologic malignancies such as acute lymphoblastic leukemia (B-ALL), various types of B-cell lymphomas, and chronic lymphocytic leukemia. The manufacturing process is patient-specific, involving leukapheresis, T-cell isolation, genetic engineering (often via viral vectors), expansion, and re-infusion.
