Autologous CD133+ hematopoietic stem cells is an investigational cell therapy developed by St. Jude Children's Research Hospital for the treatment of high-risk pediatric solid tumors and lymphomas. The therapy involves the collection of a patient's own hematopoietic stem and progenitor cells, which are then processed using the CliniMACS device to positively select for the CD133 (Prominin-1) antigen. This selection process is designed to 'purge' the graft of contaminating tumor cells—which typically do not express CD133 in the targeted pediatric cancers—while retaining the essential cells required for hematopoietic recovery. The purified CD133+ cells are re-infused into the patient following a myeloablative conditioning regimen of high-dose busulfan and melphalan to restore bone marrow function and minimize the risk of post-transplant relapse caused by graft contamination.
