Autologous CD40-activated acute lymphoblastic leukemia cell vaccine is an experimental cell-based immunotherapy developed by the Dana-Farber Cancer Institute. The vaccine is produced by collecting a patient's own malignant B-lineage acute lymphoblastic leukemia (ALL) cells and stimulating them ex vivo with CD40 ligand (CD40L). This activation triggers the leukemia cells to upregulate MHC and costimulatory molecules (such as CD80 and CD86), effectively converting the tumor cells into potent antigen-presenting cells (APCs). When these modified cells are re-administered to the patient, they are designed to present a broad array of patient-specific leukemia antigens to the endogenous immune system, thereby priming and expanding leukemia-specific T-cells to target and eliminate residual cancer cells.
