Overview

This autologous cell-based cancer vaccine, developed by Baylor College of Medicine, is designed for the treatment of B-cell chronic lymphocytic leukemia (B-CLL). The vaccine is produced by genetically modifying a patient's own malignant B-CLL cells ex vivo. It typically consists of a mixture of two cell populations: one modified to express human CD40 ligand (CD40L) and another to secrete human interleukin-2 (IL-2). CD40L expression activates the B-CLL cells, enhancing their capacity to present tumor antigens to the immune system, while IL-2 acts as a potent cytokine to stimulate and expand tumor-reactive T-cells. This dual approach aims to overcome the characteristic immune evasion of B-CLL. Clinical evaluation has included Phase 1 studies, such as the TAIL trial, which combined the vaccine with lenalidomide to further modulate the immune environment by reducing regulatory T-cell activity.
