Autologous DC-CIK and CIK therapy is an adoptive cellular immunotherapy regimen being developed by Xiaoyi Huang for the treatment of advanced solid tumors, including colorectal cancer, renal cell carcinoma, nasopharyngeal carcinoma, and lung cancer. The treatment involves the ex vivo expansion of a patient's peripheral blood mononuclear cells (PBMCs) into cytokine-induced killer (CIK) cells, which are a heterogeneous population of T cells with NK-like properties (CD3+CD56+). A portion of these cells is co-cultured with dendritic cells (DCs) that have been pulsed with multiple tumor-associated antigens to create DC-CIK cells, enhancing the specificity and potency of the immune response. The therapy is typically administered intravenously in multiple cycles, often in combination with chemotherapy, to induce MHC-unrestricted and antigen-specific tumor cell lysis through the release of perforins and granzymes and the activation of apoptotic pathways.
