Autologous Dendritic Cell Therapy (Mayo Clinic) is an investigational cell-based immunotherapy being developed for the treatment of recurrent or refractory non-Hodgkin lymphoma (NHL). The therapy utilizes a patient's own dendritic cells, which are derived from white blood cells collected via leukapheresis and matured in vitro. This therapy is administered intratumorally following cryoablation of a lymphoma lesion. The mechanism relies on cryoablation to induce immunogenic cell death, releasing tumor-associated antigens that the injected dendritic cells then capture, process, and present to T-cells. This process is intended to prime a systemic, tumor-specific immune response. In clinical evaluation (NCT03035331), this dendritic cell vaccine is used as part of a multimodal regimen that includes the PD-1 inhibitor pembrolizumab to prevent T-cell exhaustion and the pneumococcal 13-valent conjugate vaccine (Prevnar) to potentially enhance local immune activation and monitor vaccine effectiveness.
