Overview

The autologous dendritic cell vaccine developed at Cedars-Sinai Medical Center is a personalized cancer immunotherapy designed to treat malignant brain tumors, including glioblastoma and brain stem gliomas. The therapeutic process involves harvesting a patient's own monocytes from their blood, which are then differentiated into dendritic cells in a laboratory setting. These dendritic cells are 'pulsed' or loaded with specific tumor-associated antigens (TAAs), notably including cancer stem cell markers such as CD133 (Prominin-1). Once matured and re-administered to the patient via subcutaneous injection, these primed dendritic cells migrate to the lymph nodes to present the tumor antigens to T-cells. This mechanism trains the patient's immune system to recognize and destroy tumor cells and cancer stem cells expressing those antigens, aiming to target the cell populations often responsible for treatment resistance and tumor recurrence.
