Overview

This autologous dendritic cell vaccine, developed by Duke University, is an investigational cellular immunotherapy designed for the treatment of metastatic melanoma. The therapeutic process involves collecting a patient's own peripheral blood mononuclear cells via leukapheresis, which are then differentiated into dendritic cells ex vivo. These dendritic cells are pulsed with specific melanoma-associated antigens, typically gp100 and tyrosinase peptides, or sometimes autologous tumor lysate, to program them to recognize the patient's cancer cells. After maturation, the antigen-loaded dendritic cells are re-infused into the patient, where they act as professional antigen-presenting cells to activate and expand tumor-specific T-cell populations. This approach is intended to stimulate a targeted immune response against melanoma cells and is often evaluated in conjunction with systemic cytokines like interleukin-2 (aldesleukin) to further boost T-cell activity.
