Overview

Autologous dendritic cells infected with Fowlpox-CEA-TRICOM is an experimental active immunotherapy developed by Michael Morse, MD, at Duke University. This cell-based vaccine involves harvesting a patient's own dendritic cells and infecting them ex vivo with a recombinant fowlpox virus vector. This vector is engineered to express a modified version of the carcinoembryonic antigen (CEA-6D) along with a triad of costimulatory molecules (TRICOM) consisting of B7-1 (CD80), ICAM-1 (CD54), and LFA-3 (CD58). The modified dendritic cells are then re-infused into the patient to present the CEA antigen to the immune system. The inclusion of TRICOM is intended to enhance the activation and proliferation of CEA-specific T cells, thereby mounting a more robust immune response against CEA-expressing malignancies such as colorectal, pancreatic, and breast cancers.
