This autologous natural killer (NK) cell therapy is an investigational immunotherapy developed by M.D. Anderson Cancer Center for the treatment of recurrent or refractory malignant posterior fossa tumors in pediatric patients, specifically medulloblastoma, ependymoma, and atypical teratoid/rhabdoid tumors (AT/RT). The therapeutic process involves harvesting NK cells from the patient's own peripheral blood, followed by ex vivo expansion to significantly increase the cell count and enhance cytotoxic activity. These expanded NK cells are then administered directly into the brain ventricles via an intraventricular catheter (e.g., Ommaya reservoir). This localized delivery method is intended to bypass the blood-brain barrier and maximize the concentration of effector cells in the cerebrospinal fluid and tumor microenvironment while minimizing systemic side effects. The therapy is currently being evaluated in the Phase I NOAH (New Opportunity, Advancing Hope) clinical trial.
