Autologous expanded CD34+ stem cells is an investigational cell therapy developed by Imperial College London for the treatment of chronic liver insufficiency and liver cirrhosis. The therapy involves mobilizing a patient's own hematopoietic CD34+ stem cells from the bone marrow into the bloodstream using granulocyte colony-stimulating factor (G-CSF). These cells are then collected via leukapheresis, expanded ex vivo, and re-infused into the patient via the hepatic artery or portal vein. The therapeutic rationale is based on the potential of CD34+ hematopoietic stem cells to differentiate into hepatocytes and other liver cell lineages, thereby regenerating damaged hepatic tissue and reducing fibrosis. A Phase I/II dose-escalation study (NCT00655707) has evaluated the safety, tolerability, and maximum tolerated dose of this approach in adults with chronic liver insufficiency or cirrhosis.
