Overview

A cell-based therapy involving autologous hematopoietic stem cell transplantation (HSCT) for the treatment of refractory Crohn's disease. Developed by Cedars-Sinai Medical Center, the procedure involves mobilizing and collecting a patient's own peripheral blood stem cells (CD34+), followed by a lymphoablative conditioning regimen (using agents like fludarabine, cyclophosphamide, and anti-thymocyte globulin) to eliminate the aberrant, inflammatory immune system. The harvested stem cells are then reinfused to reset or reprogram the immune system to a pre-disease state, potentially inducing long-term remission in patients who have failed conventional therapies. This therapeutic approach is currently being evaluated in a Phase 1/2 open-label trial for patients aged 13-28 years with treatment-refractory Crohn's disease.
