Overview

A non-myeloablative autologous hematopoietic stem cell transplantation (HSCT) protocol developed at Northwestern University for the treatment of relapsing-remitting multiple sclerosis (RRMS). The procedure involves mobilizing a patient's own peripheral blood stem cells using cyclophosphamide and granulocyte colony-stimulating factor (G-CSF). This is followed by a conditioning regimen consisting of high-dose cyclophosphamide (200 mg/kg) and alemtuzumab (CAMPATH-1H, 20 mg) to deplete the autoreactive immune system. Finally, the harvested stem cells are re-infused to facilitate immune reconstitution and the establishment of a self-tolerant immune system. The protocol is specifically designed for patients who have failed conventional therapies, such as interferon, and aims to halt inflammatory activity and disease progression.
