Autologous hematopoietic stem cells (cryopreserved) are a patient's own multipotent stem cells, typically harvested from peripheral blood following mobilization or directly from the bone marrow, and subsequently frozen for later therapeutic use. This cellular product is the fundamental component of autologous hematopoietic stem cell transplantation (aHSCT). The primary mechanism of action is the reconstitution of the patient's hematopoietic and immune systems following high-dose, myeloablative, or lymphodepleting conditioning regimens. By reinfusing these stored cells, clinicians can administer intensive chemotherapy doses that would otherwise cause permanent bone marrow failure. Cryopreservation, usually involving dimethyl sulfoxide (DMSO), allows for the temporal separation of cell collection and the conditioning phase, facilitating treatment for malignancies such as multiple myeloma and relapsed lymphomas, as well as the 'immune resetting' required for severe autoimmune diseases like systemic sclerosis and multiple sclerosis.
