Overview

Autologous hematopoietic stem cells (HSCs) are a patient's own blood-forming cells, typically collected from the peripheral blood after mobilization or directly from the bone marrow. In the treatment of acute myeloid leukemia (AML), these cells are harvested and cryopreserved while the patient is in remission. The patient then undergoes myeloablative conditioning—intensive chemotherapy (e.g., busulfan, cyclophosphamide, etoposide) or total body irradiation—to eliminate residual leukemic cells. The autologous HSCs are subsequently thawed and re-infused intravenously to rescue the bone marrow and restore normal hematopoiesis, allowing for the use of dose-intensive therapy that would otherwise cause permanent marrow failure.
