Drug intelligence / Profile preview
Autologous hematopoietic stem cells (HSCs) are utilized as a supportive cellular therapy within the ACTION trial (Adoptive Cellular Therapy following Dose-Intensified Temozolomide in Newly-diagnosed Pediatric High-grade Gliomas), an investigational study conducted by the University of Florida. In this protocol, patients with newly-diagnosed pediatric high-grade gliomas (WHO Grade III or IV) undergo mobilized leukapheresis to collect peripheral blood stem cells (PBSCs) prior to receiving dose-intensified temozolomide (TMZ) chemotherapy. Following the intensive chemotherapy cycles, the autologous HSCs are re-infused to facilitate hematopoietic recovery and mitigate the severe myelosuppression caused by the treatment. This hematopoietic rescue is a critical component of the multi-modal treatment strategy, which also includes the administration of tumor total RNA-loaded dendritic cell (TTRNA-DC) vaccines and tumor-specific T cells (TTRNA-xALT) to stimulate an anti-tumor immune response.
