This combination therapy pairs AG-858, a personalized cancer vaccine based on heat shock protein 70 (HSP-70) protein-peptide complex derived from a patient's own tumor cells, with imatinib (Gleevec/Glivec), a BCR-ABL tyrosine kinase inhibitor. The combination aims to enhance treatment efficacy in chronic myelogenous leukemia (CML) by targeting multiple pathways simultaneously. While imatinib inhibits the BCR-ABL tyrosine kinase that drives CML progression, AG-858 is designed to stimulate an immune response against residual leukemia cells. This approach was investigated as a potential strategy to improve outcomes for patients who remained cytogenetically positive after imatinib treatment alone.
