Autologous NK cell therapy + GnRHa + add-back therapy is an experimental combination treatment regimen developed by Shenzhen People's Hospital for patients with severe (Stage III-IV) endometriosis. This therapeutic approach combines hormonal suppression with cellular immunotherapy. The regimen begins with the administration of a gonadotropin-releasing hormone agonist (GnRHa), which suppresses the hypothalamic-pituitary-ovarian axis, leading to a hypoestrogenic state that inhibits the growth of ectopic endometrial tissue. To counteract the side effects of estrogen deficiency, such as bone loss, 'reverse addition' (add-back) therapy is provided. Following this conventional treatment, patients receive infusions of autologous natural killer (NK) cells. The rationale is that patients with endometriosis often exhibit NK cell dysfunction, which prevents the effective clearance of ectopic endometrial cells; by supplementing functional autologous NK cells, the therapy aims to enhance the immune-mediated destruction of residual disease and reduce the risk of recurrence.
