Autologous regulatory T cell-depleted lymphocytes is an experimental cell therapy developed by Assistance Publique - Hôpitaux de Paris (AP-HP) for the treatment of advanced metastatic colorectal cancer, specifically targeting patients with unresectable hepatic metastases. The therapeutic process involves collecting a patient's own circulating lymphocytes via cytapheresis and performing an ex-vivo cell sorting procedure to selectively remove regulatory T cells (Tregs). Tregs are a subset of T lymphocytes that normally maintain immune tolerance but can also suppress the immune system's ability to recognize and destroy cancer cells. By depleting these suppressive cells and re-infusing the remaining lymphocyte population into the patient—typically following a lymphodepleting conditioning regimen of cyclophosphamide and fludarabine—the therapy aims to enhance the effector immune response against the tumor.
