Overview

Autologous RNA-electroporated mature dendritic cells expressing MAGE-3 + survivin + BCMA is an investigational, patient-specific dendritic cell (DC)-based cancer vaccine developed by the Universitair Medisch Centrum Sint Radboud (Radboud UMC). The vaccine consists of autologous mature monocyte-derived dendritic cells electroporated with mRNA encoding three tumor-associated antigens (TAAs) highly expressed on myeloma cells: MAGE-3 (a cancer-testis antigen), survivin (an apoptosis inhibitor), and BCMA (B-cell maturation antigen). Additionally, the cells are pulsed with keyhole limpet hemocyanin (KLH) to serve as an immunological adjuvant. Designed to treat multiple myeloma patients with minimal residual disease (MRD) following intensive chemotherapy and autologous stem cell transplantation (ASCT), the vaccine aims to stimulate antigen-specific T-cell responses to eliminate residual cancer cells. In a Phase 1 clinical trial, the vaccine was well tolerated and demonstrated the capability to induce tumor antigen-specific T-cell responses.
