Overview

A combination immunotherapy composed of an autologous tumor cell vaccine and a CpG oligodeoxynucleotide adjuvant. The therapy is produced by isolating and processing a patient's own (autologous) tumor cells into a whole-cell vaccine formulation in vitro, then combining these cells with unmethylated cytosine-phosphate-guanine (CpG) oligodeoxynucleotides—synthetic DNA sequences that mimic bacterial DNA and act as immune stimulants. The resulting product is administered back to the patient. The mechanism of action involves two synergistic components: - The autologous tumor cells provide a broad array of patient-specific antigens. - The CpG oligodeoxynucleotides activate antigen-presenting cells (APCs), such as dendritic cells and macrophages, via Toll-like receptor 9 (TLR9), enhancing uptake and presentation of the tumor antigens. This leads to robust activation of both CD4+ helper T-cells and CD8+ cytotoxic T-cells against the cancer[2][3][5][6]. This approach has been investigated primarily for hematologic malignancies such as mantle cell lymphoma, but also in solid tumors including melanoma and colorectal cancer[2][3]. Clinical trials have demonstrated safety, feasibility, induction of anti-tumor immune responses, reduction in minimal residual disease after stem-cell transplantation for lymphoma patients, and some evidence for improved clinical outcomes[2][3].
