Overview

Autologous tumor lysate is a personalized biological product derived from a patient's own tumor tissue, used as an antigenic source in cancer immunotherapy. In the context of the Phase I clinical trial led by Edward Ziga (NCT03615404), the lysate is prepared from refractory pediatric central nervous system (CNS) tumors. It is administered intradermally following a dendritic cell (DC) vaccine and in conjunction with topical imiquimod. The lysate provides a comprehensive set of tumor-associated antigens (TAAs) unique to the patient's malignancy, which are then processed and presented by dendritic cells to prime a specific T-cell-mediated anti-tumor immune response. This approach aims to overcome the immunosuppressive environment of brain tumors and improve survival in children with refractory disease.
