This autologous dendritic cell-based immunotherapy is developed by the Fondazione I.R.C.C.S. Istituto Neurologico Carlo Besta for the treatment of glioblastoma multiforme (GBM). The vaccine is produced by isolating monocytes from a patient's peripheral blood, which are then differentiated into mature dendritic cells (mDCs) using a specific cytokine cocktail including IL-4, GM-CSF, TNF-α, PGE2, IL-1β, and IL-6. These mDCs are subsequently pulsed with autologous tumor lysate derived from the patient's own surgically resected tumor tissue. This process allows the dendritic cells to present a broad spectrum of patient-specific tumor antigens to the immune system. Once administered, the vaccine aims to prime and activate T cells to recognize and eliminate residual glioblastoma cells. It has been evaluated in clinical trials for both newly diagnosed (DENDR1) and recurrent (DENDR2) glioblastoma, often in combination with standard-of-care treatments like the Stupp protocol.
