AV-MEL-1 + anti-PD-1 is an investigational combination therapy for metastatic melanoma consisting of a personalized autologous dendritic cell vaccine (AV-MEL-1) loaded with patient-specific tumor antigens and an anti-PD-1 monoclonal antibody (either pembrolizumab or nivolumab). AV-MEL-1 is manufactured by harvesting tumor tissue from each patient, establishing a short-term cell line, and loading autologous dendritic cells—derived from the patient’s PBMCs via leukapheresis—with these tumor antigens. Anti-PD-1 antibodies block the PD-1 receptor on T cells, preventing interaction with PD-L1/PD-L2 on tumor cells, thereby inhibiting immune checkpoint-mediated T cell suppression and reactivating anti-tumor immune responses. The combination aims to enhance and sustain anti-melanoma immunity by combining personalized antigen presentation with immune checkpoint inhibition[1][3][5][6].
