A combination of **avanafil** and **desipramine** would include both mechanisms and indications of these two small molecule drugs. **Avanafil** is a phosphodiesterase 5 (PDE5) inhibitor used for erectile dysfunction, acting by enhancing nitric oxide-mediated vasodilation in the corpus cavernosum. **Desipramine** is a tricyclic antidepressant primarily inhibiting norepinephrine reuptake, used in major depressive disorder. No fixed-dose combination of avanafil and desipramine is currently marketed, approved, or under development. When coadministered, avanafil (a weak CYP2D6 inhibitor) may slightly increase plasma concentrations of desipramine (a CYP2D6 substrate), but this interaction is considered minor[1][4].
