AVE1642 + fulvestrant is a combination therapy consisting of **AVE1642**, a humanized monoclonal antibody targeting **insulin-like growth factor 1 receptor (IGF-1R)**, and **fulvestrant**, a selective estrogen receptor degrader (SERD). AVE1642 blocks IGF-1R, a pathway implicated in cancer cell proliferation and resistance to therapy, while fulvestrant degrades estrogen receptors, leading to inhibition of estrogen signaling in hormone-dependent cancers such as breast cancer. The combination was investigated as a treatment for advanced hormone-dependent breast cancer in postmenopausal women. AVE1642 was developed by Sanofi/ImmunoGen and binds IGF-1R with high affinity, down-regulating its downstream signaling pathways. The main modality is monoclonal antibody for AVE1642, and small molecule hormonal therapy for fulvestrant[7][8][4].
