Overview

Avelumab + bempegaldesleukin + enzalutamide is an investigational combination therapy evaluated in clinical trials, primarily for metastatic castration-resistant prostate cancer (mCRPC) after progression on abiraterone. The regimen includes: - **Avelumab**: a human anti-PD-L1 monoclonal antibody that blocks the interaction between PD-L1 and PD-1, enhancing T-cell anti-tumor activity. - **Bempegaldesleukin (NKTR-214)**: a PEGylated interleukin-2 (IL-2) cytokine therapy that preferentially activates and expands CD8+ T cells and NK cells within the tumor microenvironment, boosting immune response against cancer cells. - **Enzalutamide**: a competitive androgen receptor inhibitor that blocks the effect of P450 androgens (testosterone) on prostate cancer cells, preventing their growth and survival. The combination is intended to leverage both immunotherapeutic and hormonal mechanisms for synergistic effects in mCRPC.
