**Avelumab + binimetinib + talazoparib** is a proposed investigational combination regimen involving three anticancer agents with distinct mechanisms of action: avelumab, a monoclonal antibody targeting programmed death-ligand 1 (PD-L1); binimetinib, a small molecule inhibitor of mitogen-activated protein kinase kinase (MEK1/2); and talazoparib, a poly(ADP-ribose) polymerase (PARP) inhibitor. This triplet was designed for potentially additive or synergistic antitumor effects in various cancers, particularly those with DNA damage response deficiencies. The combination entered early-phase clinical trials in metastatic pancreatic ductal adenocarcinoma and other advanced solid tumors, but the specific triplet (all three drugs together) did not proceed beyond phase 1 dose-finding because of safety and feasibility concerns. Only doublet combinations (avelumab + binimetinib and binimetinib + talazoparib) were tested in human subjects, not the full triplet combination[1][3][5]. Each component is approved for various cancer indications as monotherapy or in other combinations.
