Overview

This refers to a combination treatment regimen being evaluated in the TRIUMPH-B01 Phase II clinical trial for muscle-invasive bladder cancer (MIBC). The regimen consists of the PD-L1 inhibitor avelumab (Bavencio) administered alongside platinum-based chemotherapy (either gemcitabine/cisplatin or dose-dense MVAC). This pharmacological combination is part of a tetra-modality bladder preservation strategy that also includes maximal transurethral resection of bladder tumor (TURBT) and radiation therapy. Avelumab acts as an immune checkpoint inhibitor to enhance the body's anti-tumor immune response by blocking the interaction between PD-L1 and PD-1, while the chemotherapy provides systemic cytotoxic effects and acts as a radiosensitizer. The study is led by the American University of Beirut Medical Center.
