Overview

**Avelumab + cisplatin** is an investigational drug combination used as neoadjuvant therapy primarily for muscle-invasive bladder cancer (MIBC). Avelumab is a human anti–PD-L1 monoclonal antibody (immune checkpoint inhibitor) that restores T cell–mediated antitumor immunity by blocking PD-L1 interaction with PD-1. Cisplatin is a platinum-based cytotoxic chemotherapy that forms DNA crosslinks leading to apoptosis. Combining these agents leverages the immune-stimulating effects of chemotherapy with checkpoint inhibition for potentially synergistic anticancer effects. This combination is under investigation in several clinical trials, such as the phase 2 AURA trial. The regimen has not received standalone regulatory approval for any indication, but both drugs are approved individually and in related combinations.
