Overview

This is a **combination regimen** comprised of three immunotherapies: - **avelumab**: a fully human IgG1 monoclonal antibody that targets programmed death-ligand 1 (PD-L1), blocking its interaction with the PD-1 receptor, thereby enhancing antitumor T-cell activity through immune checkpoint inhibition. - **CMP-001**: a CpG-A oligodeoxynucleotide packaged in a virus-like particle, acting as a Toll-like receptor 9 (TLR9) agonist; it stimulates plasmacytoid dendritic cells and induces interferon production, designed to trigger a robust immune response against tumor cells. - **utomilumab**: also known as PF-05082566, is a fully human agonistic monoclonal antibody that targets 4-1BB (CD137), a costimulatory molecule on immune cells; by engaging this receptor, utomilumab enhances T-cell proliferation and activation, strengthening cellular immune responses against cancer. This triplet regimen is being investigated as an immunotherapy combination for patients with advanced malignancies, including but not limited to solid tumors and certain lymphomas. The rationale is to simultaneously block inhibitory signals (avelumab), stimulate innate immune responses (CMP-001), and enhance T-cell costimulation (utomilumab) to promote a synergistic antitumor effect.
