Overview

This drug is a combination of **avelumab**, a fully human anti-PD-L1 monoclonal antibody, and **M6223**, a fully human monoclonal antibody targeting TIGIT (T cell immunoreceptor with Ig and ITIM domains). \n- **Avelumab** acts by blocking PD-L1, preventing the inhibition of immune responses on cancer cells, thereby reactivating T cell-mediated antitumor immunity. \n- **M6223** blocks TIGIT, releasing immune inhibitory "brakes" on T cells and natural killer (NK) cells, and thus enhances antitumor activity. \nThe combination is being evaluated as a **maintenance immunotherapy for advanced urothelial carcinoma (bladder cancer)** following response or stable disease after platinum-based chemotherapy, aiming to improve outcomes by dual checkpoint inhibition. \nClinical evidence from early-phase trials indicates the combination can enhance natural killer cell activation and provide additive immunomodulatory effects, with development currently at the phase II stage in the JAVELIN Bladder Medley trial[NCT05327530][1][2][3][5][7].
