Overview

**Avelumab + utomilumab** is a combination of two monoclonal antibodies used in investigational cancer therapy, typically in advanced or relapsed/refractory malignancies. **Avelumab** is a fully human IgG1 monoclonal antibody targeting PD-L1, functioning as a checkpoint inhibitor by blocking PD-L1 interaction with PD-1, thereby enhancing T-cell mediated antitumor responses and also mediating antibody-dependent cell-mediated cytotoxicity (ADCC)[2][4][8]. **Utomilumab** is a fully human IgG2 monoclonal antibody that acts as an agonist of 4-1BB (CD137), stimulating co-stimulatory signals for T-cell proliferation and activation to further augment immune response against tumors[1][7][9]. This combination is under clinical investigation mainly for aggressive lymphomas and solid tumors, aiming to synergistically activate the immune system for improved antitumor activity.
