AvidinOX is a proprietary platform technology developed by Kiran Biotech, utilizing a chemically oxidized form of avidin. When injected into a target tissue, such as a tumor, AvidinOX forms covalent bonds with local proteins, creating a stable artificial receptor that persists for weeks. This platform is designed to capture and concentrate biotinylated therapeutic agents at the site of injection. In the context of HER2-positive tumors, the platform is used to deliver and localize the HER2-targeted monoclonal antibodies trastuzumab and pertuzumab. Trastuzumab binds to domain IV of the HER2 receptor, while pertuzumab binds to domain II, together providing a comprehensive blockade of HER2 signaling and dimerization. The combination aims to enhance the anti-tumor activity of these antibodies by maintaining high local concentrations within the tumor microenvironment while potentially reducing systemic exposure.
