The HER2 alphavaccine (AVX701) is an RNA-based cancer vaccine developed by AlphaVax for the treatment of HER2-positive breast cancer. It utilizes the Venezuelan equine encephalitis (VEE) virus-like replicon particle (VRP) platform to deliver a self-replicating RNA replicon encoding the HER2/neu antigen. The vaccine is designed to target dendritic cells, where the RNA is translated into the HER2 protein, thereby stimulating both cellular (CD8+ T cell) and humoral (antibody) immune responses against HER2-overexpressing tumor cells. Clinical studies have indicated that AVX701 can break immune tolerance to HER2 and may improve progression-free survival in patients with advanced breast cancer, particularly when used in combination with other immunotherapies like checkpoint inhibitors.
