**AVZO-021 + sacituzumab govitecan-hziy** is an investigational combination therapy currently in clinical trials for the treatment of advanced solid tumors, with a focus on hormone receptor-positive (HR+), human epidermal growth factor receptor 2-negative (HER2-) metastatic breast cancer. AVZO-021 is a highly selective, oral cyclin-dependent kinase 2 (CDK2) inhibitor. Sacituzumab govitecan-hziy is a Trop-2-directed antibody-drug conjugate (ADC), consisting of a humanized monoclonal antibody targeting Trop-2 conjugated to the cytotoxic agent SN-38, a topoisomerase I inhibitor. The rationale for the combination is to enhance anti-tumor activity by simultaneously inhibiting cell cycle progression and targeting Trop-2-positive cancer cells with cytotoxic payload. The combination is being evaluated in an ongoing Phase 1/2 clinical trial[2][4].
