Overview
AVZO-023 + fulvestrant is an oral combination therapy being investigated for the treatment of advanced or metastatic hormone receptor-positive (HR+)/human epidermal growth factor receptor 2-negative (HER2-) breast cancer and select advanced solid tumors. AVZO-023 is a highly potent, selective cyclin-dependent kinase 4 (CDK4) inhibitor developed to improve efficacy and minimize toxicity by targeting a key driver of HR+/HER2- breast cancer. Fulvestrant is a selective estrogen receptor degrader (SERD), an antiestrogen agent that binds the estrogen receptor, leading to its degradation and inhibition of downstream signaling. The combination aims to arrest tumor growth through dual cell cycle and hormone receptor pathway inhibition. AVZO-023 is developed by Avenzo Therapeutics, which holds worldwide rights excluding Greater China, secured from Allorion Therapeutics. The combination is being evaluated in a Phase 1/2 clinical trial for safety, tolerability, pharmacokinetics, pharmacodynamics, and antitumor activity[1][2][5][6][8][9][10].
