Overview

This combination therapy consists of axatilimab, a humanized IgG4 monoclonal antibody targeting the colony-stimulating factor 1 receptor (CSF-1R), and nivolumab, a programmed death receptor-1 (PD-1) blocking antibody. The combination is currently being evaluated in a Phase II proof-of-concept clinical trial (NCT05723055) sponsored by the University of Utah for the treatment of relapsed or refractory classical Hodgkin lymphoma. Axatilimab works by binding to CSF-1R expressed on monocytes and macrophages, thereby reducing the levels of proinflammatory and profibrotic cells that contribute to the tumor microenvironment. Nivolumab acts as a checkpoint inhibitor, enhancing T-cell mediated anti-tumor responses. This investigational regimen is specifically targeted at patients who have progressed or shown suboptimal responses to prior anti-PD-1/PD-L1 therapies.
