Axatilimab + pembrolizumab is a combination therapy that pairs two distinct immunotherapeutic agents. Axatilimab is a high-affinity monoclonal antibody that targets colony stimulating factor 1 receptor (CSF-1R), inhibiting CSF-1R signaling and restraining macrophage development. This reduces circulating levels of pro-inflammatory and pro-fibrotic monocytes and macrophages, ultimately resulting in reduced activity of pathogenic macrophages in tissues. Pembrolizumab is a programmed cell death protein 1 (PD-1) inhibitor that blocks the interaction between PD-1 and its ligands PD-L1 and PD-L2, releasing PD-1 pathway-mediated inhibition of the immune response and improving anti-tumor immune response. The combination is being investigated for the treatment of Hodgkin lymphoma.
