Axitinib + cisplatin + pemetrexed is a combination regimen of three drugs used in the treatment of certain cancers. Axitinib is a small molecule tyrosine kinase inhibitor that selectively inhibits vascular endothelial growth factor receptors (VEGFR) 1, 2, and 3 to reduce angiogenesis. Cisplatin is a platinum-based chemotherapy agent that causes DNA crosslinking and apoptosis in rapidly dividing cells. Pemetrexed is an antifolate chemotherapeutic agent that disrupts folate-dependent metabolic processes essential for cell replication. This combination has been studied primarily in advanced non-squamous non-small-cell lung cancer (NSCLC) and malignant pleural mesothelioma. Clinical trials have shown the regimen to be generally well tolerated but without significant improvement in progression-free or overall survival compared to standard chemotherapy alone[1][2][6].
