axitinib + gemcitabine

**Axitinib + gemcitabine** is a combination therapy comprised of axitinib, an oral small molecule inhibitor of vascular endothelial growth factor receptors 1, 2, and 3 (VEGFR-1, -2, -3), and gemcitabine, a nucleoside analog chemotherapeutic agent that inhibits DNA synthesis. Axitinib targets angiogenesis by blocking VEGFR tyrosine kinases, while gemcitabine acts as an antimetabolite interfering with DNA replication. This combination has been investigated in phase I–III trials for advanced pancreatic cancer. Clinical studies show the combination is generally well tolerated, without significant pharmacokinetic interactions, but does not provide a survival benefit over gemcitabine alone in advanced pancreatic cancer[1][2][3][4][5].
